  • Cell Concentration (cells/mL) = (Cavg × D × 10,000)
  • Total Cells in Sample = Cell Concentration × Vsample
  • Viability (%) = (Lcells / Tcells) × 100

Where:

  • Cavg: Average number of cells counted per large square (1mm² area, 0.1mm depth).
  • D: Dilution Factor (e.g., 1 for undiluted, 2 for 1:1 dilution, 10 for 1:9 dilution).
  • Vsample: Original total sample volume (in mL).
  • Lcells: Number of live cells counted (for viability).
  • Tcells: Total number of cells counted (live + dead, for viability).
